Von besonderer Relevanz ist die examine persistenter organischer Schadstoffen (POPs), da sie ein hohes toxisches strength haben, schlecht abbaubar sind und sich daher intestine in biologischen Organismen anreichern können. Dabei konnte mit konventionellen Methoden meist nur der Gesamtgehalt von Schadstoffen in der Matrix analysiert werden. Auch die severe Spurenanalyse von Substanzen struggle bei herkömmlichen Verfahren oft problematisch. Der Fokus bei diesen Analysemethoden, die hauptsächlich auf einer Extraktion mittels polysiloxanmodifizierter Festphasen basieren, ist vor allem seit einigen Jahren auf einen reduzierten Lösungsmittelverbrauch und auf möglichst weit reichende Automatisierung gerichtet. Dabei wird als Extraktionsmaterial am häufigsten Polydimethylsiloxan (PDMS) verwendet. Die chemischen Vorgänge dieser Analysen verlaufen über eine Absorption der Analyten in das Polymermaterial. Im Gegensatz zu den Adsorptionsprozessen, wie bei einer Festphasenextraktion (SPE), ist der Absorptionsprozess schwächer. Daher sind die Bedingungen der Rückextraktion bzw. der Desorption der Analyten milder. Zur anschließenden examine der Schadstoffe stehen verschiedene Verfahren zur Verfügung. Hier sei vor allem die Gaschromatographie (GC) genannt, die, gekoppelt mit einem leistungsfähigen Detektor wie dem Massenspektrometer (MS), zu den empfindlichsten und gleichzeitig selektivsten Analysenverfahren zählt. Für die Anwendung eines GC/MS – platforms müssen sich die zu analysierenden Verbindungen unzersetzt oder reproduzierbar zersetzt im Injektor des GC verdampfen lassen. Die Proben können mit unterschiedlichen Methoden für die instrumentelle GC/MS – Bestimmung vorbereitet werden. Bei der Thermodesorption werden die mit dem Analyten beladenen Silikonstäbe in ein leeres Thermodesorptionsröhrchen transferiert und die Analyten durch hohe Temperaturen ausgetrieben. Dies ist eine komplett lösungsmittelfreie Anreicherungstechnik. Bei der anderen Variante wird eine Rückextraktion mit kleinen Lösungsmittelmengen (ca. a hundred µL – 500 µL) durchgeführt. Für beide aufgeführten Methoden ist die GC/MS als Analyseverfahren ideal.
